Avmapki Fakzynja Co-Pack (avutometinib plus defactinib) is manufactured by Verastem Oncology and was approved by the US FDA in 2025 for adult patients with KRAS-mutant recurrent low-grade serous ovarian cancer (LGSOC). Both halves of the co-pack are oral small molecules taken on a structured cycle.
The regimen targets the RAS/MAPK pathway from two angles. Avutometinib is a RAF/MEK clamp that binds the RAF-MEK complex and blocks the feedback reactivation of MEK seen with single-agent MEK inhibition. Defactinib is a focal adhesion kinase (FAK) inhibitor that suppresses an adaptive resistance node. The two molecules are co-packaged so the dosing schedule for each half is paired correctly at the patient level.
