About Wayrilz
Wayrilz (rilzabrutinib) is an oral reversible covalent Bruton's tyrosine kinase (BTK) inhibitor from Sanofi, originally developed by Principia Biopharma. It is indicated for adults with persistent or chronic immune thrombocytopenia (ITP) who have had an insufficient response to a previous treatment. It earned US FDA approval in 2025.
Rilzabrutinib's reversible binding profile distinguishes it from earlier irreversible BTK inhibitors and supports a dual mechanism in ITP, modulating both autoantibody-driven platelet destruction (via B-cell and Fc-receptor signalling) and inflammatory macrophage activity, without targeting platelets directly.
